• 繁殖方式之一：交配繁殖（卵生）。巨蜥约于9月产卵。刚成熟的雌蜥只能产4-6枚卵，每隔2-3天产一次，一窝卵平均有20颗卵，孵卵期为7个月，雌性会将卵产在地底或树穴，保护它们。十岁左右，进入产卵旺期，每次产下二十几枚，将卵埋在沙窝里，靠太阳辐射的自然温度孵卵，八个月后，幼蜥才破壳而出。刚出壳时，小蜥大小如同我们饲养的家鹅。它能活50年左右。
• 繁殖方式之二：也可以孤雌繁殖（不需要雄性即可繁殖后代），即卵不经过受精也能发育成正常的新个体。在英国的切斯特郡立动物园在2006年5月期间，一只单独饲养的雌性科莫多龙flora居然发生了孤雌生殖的奇特现象——也算是称得上是孤雌生殖现在在蜥蜴中的最高等的表现。
